Using soybean isoflavone glycosides as raw material, the isoflavone aglycones were prepared by enzymatic hydrolysis. With the hydrolysis rate and aglycones yield as the evaluation indexes, the β-glucosidase, β-galactosidase and cellulase from different sources were screened to determine the optimal hydrolysis enzyme. The enzyme dosage, substrate mass concentration, enzymolysis temperature, pH and enzymolysis time were optimized by single factor experiment. The results showed that the optimal enzymolysis conditions were obtained as follows: β-glucosidase(300 U/g), enzyme dosage 7%, substrate mass concentration 1.6 mg/mL, enzymolysis temperature 56 ℃, pH 4.8, and enzymolysis time 6 h. Under these conditions, the hydrolysis rate of soybean isoflavone glycosides and aglycones yield reached 96.84% and 99.74%, respectively. |
