Abstract: |
The fatty acid composition of phospholipids in seawater fish Lateolabrax japonicus muscle was studied. The method for analyzing the phospholipids composition by high performance liquid chromatography-evaporative light scattering detector(HPLC-ELSD)was established. Chromolith Performance-Si type positive phase silica gel column(100 mm × 4.6 mm)was used as separation column, and n-hexane-isopropanol-acetic acid was used as mobile phase system with three element gradient elution and flow rate 1.5 mL/min. Each phospholipids component was qualified by retention time and quantified by peak area.The results indicated that the content of unsaturated fatty acids in Lateolabrax japonicus muscle phospholipids was 62.59%, and the total content of EPA and DHA was 30.86%. Each phospholipids component was separated well by HPLC-ELSD.The method had high precision, and the coefficient of variation was less than 3%. The average recovery rate was in the range of 86.00%-105.00%, and the relative standard deviation was less than 3.50%. The method was accurate and efficient for the determination of phospholipids composition in Lateolabrax japonicus muscle, and was suitable for the determination of phospholipids in seawater fish. Six kinds of phospholipids components were detected in Lateolabrax japonicus muscle. Among them, the content of phosphatidylethanolamine (PE) was the highest, followed by phosphatidylcholines(PC) and phosphatidylinositol(PI). |
