A method for rapid determination of sodium copper chlorophyllin in virgin olive oil by liquid-liquid extraction and ultra-high performance liquid chromatography (UPLC)was established. Virgin olive oil was extracted by 80% ethanol, then injected to ultra-high performance liquid chromatography, and determined under the conditions of SVEA C18 column (50 mm×1.0 mm, 1.7 μm) with isocratic elution at 30 ℃, acetonitrile-water (volume ratio 95∶ 5) as mobile phase, flow rate 0.3 mL/min, injection volume 2.0 μL, and detection wavelength 405 nm,the sodium copper chlorophyllin could be effectively separated from virgin olive oil,and the entire analysis process took only 6 min. The results of the methodological examination showed that the detection limit and quantitative limit of the method were 0.20 mg/L and 0.5 mg/L respectively, the recovery rate of sodium copper chlorophyllin was between 84.00% and 97.90%, and the method had better reproducibility, precision and stability,and could be used for the determination of sodium copper chlorophyllin in virgin olive oil. |
