Separation and purification of dilinoleoylphosphatidylcholine from soybean lecithin |

Abstract: |
Lecithin (PC) was obtained from soybean powder phospholipids by purification with 95% ethanol solution, and then dilinoleoylphosphatidylcholine (DLPC) was separated and purified by reverse phase chromatography preparation system. The effects of eluent type, flow rate of eluent and loading amount on the separation and purification of DLPC were investigated. The results showed that the purity of DLPC was 91.2% and the yield was 21.5% when methanol solution was used as eluent with a volume fraction of 95%, the flow rate of elution was 28 mL/min and the loading amount was 0.5 g (based on 40 g C18 column material). The purity of DLPC was 92.6% and the yield was 20.1% when ethanol solution was used as eluent with a volume fraction of 80%, the flow rate of elution was 28 mL/min, and the loading amount was 0.75 g(based on 40 g C18 column material). In terms of the purity of DLPC, ethanol solution as the eluent is superior to methanol solution. |
