Preparation of phytosterol oleate and its application in astaxanthin-loaded liposomes

Abstract:
      In order to explore the feasibility of preparing liposomes by phytosterol esters, using phytosterol and oleic acid as materials, phytosterol oleate was prepared, separated and structure characterized by infrared spectroscopy and nuclear magnetic resonance. The complex liposomes of astaxanthin-phytosterol oleate were prepared by using soy phospholipids as the main membrane material by a thin-film ultrasound method,and its particle size and distribution, Zeta potential, encapsulation efficiency, and morphology were studied. The results showed the purity of phytosterol oleate was (96.51±0.93)% after purification. The prepared astaxanthin-phytosterol oleate complex liposomes were nearly spherical with regular shape and good dispersion, and the average particle size was (124.0±12.5) nm with the polydispersity index (PDI) of 0.37±0.01, the average Zeta potential was (-33.3±1.6) mV and the encapsulation efficiency was (93.95±0.92)%. In conclusion, using phytosterol oleate for loading astaxanthin, the encapsulation rate is high and the prepared complex liposomes are stable and uniformly distributed, and the phytosterol oleate can be applied to the construction of liposomes loaded with lipid-soluble bioactive substances.
