Protective effect of odd-chain fatty acids on lipopolysaccharide- induced inflammation in Caco-2 cells

Abstract:
      In order to explore the health effects of odd-chain fatty acids (OCFA), serum OCFA levels in the healthy individuals and colon cancer patients were measured by gas chromatography-mass spectrometry (GC-MS) to investigate the differences in the distribution characteristics, and the protective effects of OCFA on inflammation by using the lipopolysaccharide (LPS)-induced Caco-2 cell inflammation model were studied, including the changes in cell activity, pro-inflammatory factor gene expression and fatty acid composition. The results showed that the levels of two main OCFA (C15∶ 0 and C17∶ 0) in healthy individuals were significantly higher than that in colon cancer patients.The levels of C15∶ 0 and C17∶ 0 in male patients and patients over the age of 59 were significantly lower. C15∶ 0 and C17∶ 0 had no effect on cell activity. C15∶ 0 could significantly inhibit the expression of pro-inflammatory factors TNF-α, IL-8 and iNOS in Caco-2 cells with LPS stimulation, C17∶ 0 could significantly reduce the expression of IL-6, TNF-α, IL-8 and iNOS. Two main OCFA could play a similar role in alleviating inflammatory stimulation as n-3 PUFA. Two main OCFA could be easily incorporated by Caco-2 cells into C17∶ 0 and C17∶ 1 in Caco-2. In conclusion, OCFA may have a potential preventive effect on inflammation and intestinal health by regulating the expression of pro-inflammatory cytokines.
